The emergence of antibiotic-resistant Mycobacterium tuberculosis (Mtb) poses a significant global health challenge, undermining efforts to control and eradicate tuberculosis (TB). This study provides a comprehensive overview of the mechanisms underlying antibiotic resistance in Mtb, including genetic mutations, efflux pumps, and biofilm formation. Key findings indicate that mutations in the rpoB gene confer resistance to rifampicin, while mutations in the katG and inhA genes are associated with isoniazid resistance. Mutations in the gyrA and gyrB genes have been implicated in fluoroquinolone resistance, and alterations in the rpsL and rrs genes are linked to streptomycin resistance. Additionally, the role of efflux pumps in multi-drug resistance (MDR-TB) is explored. The study also identifies biofilm formation as a significant factor in persistent infection and drug tolerance. Addressing antibiotic resistance in Mtb requires a multifaceted approach, integrating enhanced diagnostic tools, new drug development, and robust public health initiatives to curb the spread of resistant strains and improve patient outcomes. |
